The use of oxidoreductases (EC1) in non-conventional reaction media has been increasingly explored. In particular, deep eutectic solvents (DESs) have emerged as a novel class of solvents. Herein, an in-depth study of bioreduction with an alcohol dehydrogenase (ADH) in the DES glyceline is presented. The activity and stability of ADH in mixtures of glyceline/water with varying water contents were measured. Furthermore, the thermodynamic water activity and viscosity of mixtures of glyceline/water have been determined. For a better understanding of the observations, molecular dynamics simulations were performed to quantify the molecular flexibility, hydration layer, and intraprotein hydrogen bonds of ADH. The behavior of the enzyme in DESs follows the classic dependence of water activity (aW) in non-conventional media. At low aW values (<0.2), ADH does not show any activity; at higher aW values, the activity was still lower than that in pure water due to the high viscosities of the DES. Physical exercise is known to influence hormonal mediators of appetite, but the effect of short-term maximal intensity exercise on plasma levels of appetite hormones and cytokines has been little studied. We investigated the effect of a 30 s Wingate Test, followed by a postprandial period, on appetite sensations, food intake, and appetite hormones. Twenty-six physically active young males rated their subjective feelings of hunger, prospective food consumption, and fatigue on visual analogue scales at baseline, after exercise was completed, and during the postprandial period. Blood samples were obtained for the measurement of nesfatin-1, ghrelin, leptin, insulin, pancreatic polypeptide (PP), human growth factor (hGH) and cytokine interleukin-6 (IL-6), irisin and plasma lactate concentrations, at 30 min before exercise, immediately (210 s) after exercise, and 30 min following a meal and at corresponding times in control sedentary males without ad libitum meal intake, respectively. Appetite perceptions and food intake were decreased in response to exercise. Plasma levels of irisin, IL-6, lactate, nesfatin-1 and ghrelin was increased after exercise and then it was returned to postprandial/control period in both groups. A significant rise in plasma insulin, hGH and PP levels after exercise was observed while meal intake potentiated this response. In conclusion, an acute short-term fatiguing exercise can transiently suppress hunger sensations and food intake in humans. We postulate that this physiological response involves exercise-induced alterations in plasma hormones and the release of myokines such as irisin and IL-6, and supports the notion of existence of the skeletal muscle–brain–gut axis. Nevertheless, the detailed relationship between acute exercise releasing myokines, appetite sensations and impairment of this axis leading to several diseases should be further examined. 相似文献

An experimental study of wind farm blockage has been performed to quantify the velocity decrease that the first row of a wind farm experiences due to the presence of the other turbines downstream. The general perception has been that turbines downstream of the first row are only influenced by the wakes from upstream turbines without any upstream effect. In the present study, an attempt is made to demonstrate the existence of a two‐way coupling between individual turbines and turbines in the wind farm. Several staggered layouts were tested in the wind tunnel experiments by changing the spacing between rows, spacing between turbines in the rows, and the amount of wind turbines involved. The experiments focused on turbines located in the center of the first row as well as the two turbines located in the row edges, usually believed to experience a speedup. The present results show that no speedup is present and that all the turbines in the first row are subjected to a reduced wind speed. This phenomenon has been considered to be due to “global blockage.” An empirical correlation formula between spacing, number of rows, and velocity decrease is proposed to quantify such effect for the center turbine as well as for the turbines at the edges. 相似文献

The demand for general reduction of the energy consumption in civil engineering leads to more frequent use of insulating materials with air gaps or cavities. Heat transfer through a constructional part can be decreased by adding an air gap and low emissivity reflective foils to the structure. In the first part of this paper, the impacts of cavity thickness and inner surface emissivity on combined conduction, convection and radiation heat transfer was experimentally explored in the case of constructional part with a horizontal cavity subjected to constant downward heat flux. The heat flow meter Netzsch HFM 436 Lambda was used for steady-state measurements. Results suggest that the studied parameters seriously affect the combined heat transfer in the composed structure. In the second part the paper reports the numerical study of two-dimensional conjugate heat transfer in closed horizontal cavity having air as the intervening medium. Numerical models validated by related experimental results were performed to further investigate the effect of radiation heat transfer. It was found that in general, the total heat flux through the composed structure decreases with increasing air cavity thickness, which is significant especially when low emissivity inner surfaces are taking into account. The direction of heat flow (downward or upward heat flow) has a significant impact on the convection heat transfer. An important contribution from the present work is the analysis of the optimal thickness of the cavity at different boundary conditions. The optimal thickness of the enclosure with low emissivity surfaces is 16 mm when subjected to upward heat flux. 相似文献

ABSTRACT Additive manufacturing of Al-alloys allows the production of components with a complicated structured shape, geometry composed by lattice structures, internal cooling, etc. The portfolio of Al-alloys for metal additive manufacturing is still under development and is strongly limited, compared to the conventional technology. The alloy AlSi9Cu3 is used in many applications, but its processing details are still missing. The main aim of this paper is to describe the laser process parameters for AlSi9Cu3, processed by SLM technology and manufactured from two powders of different shapes and particle sizes. The tested process parameters were laser power, laser speed, and hatch distance in the range of 100–400?W, 200–1500?mm?·?s?1 and 90–150?µm. These were tested using a single-track and cube test. Chagas disease is becoming a worldwide problem; it is currently estimated that over six million people are infected. The two drugs in current use, benznidazole and nifurtimox, require long treatment regimens, show limited efficacy in the chronic phase of infection, and are known to cause adverse effects. Phenotypic screening of an in-house library led to the identification of 2,2′-methylenebis(5-(4-bromophenyl)-4,4-dimethyl-2,4-dihydro-3H-pyrazol-3-one), a phenyldihydropyrazolone dimer, which shows an in vitro pIC50 value of 5.4 against Trypanosoma cruzi. Initial optimization was done by varying substituents of the phenyl ring, after which attempts were made to replace the phenyl ring. Finally, the linker between the dimer units was varied, ultimately leading to 2,2′-methylenebis(5-(3-bromo-4-methoxyphenyl)-4,4-dimethyl-2,4-dihydro-3H-pyrazol-3-one (NPD-0228) as the most potent analogue. NPD-0228 has an in vitro pIC50 value of 6.4 against intracellular amastigotes of T. cruzi and no apparent toxicity against the human MRC-5 cell line and murine cardiac cells. 相似文献

Gasoline engine emissions have been classified as possibly carcinogenic to humans and represent a significant health risk. In this study, we used MucilAir™, a three-dimensional (3D) model of the human airway, and BEAS-2B, cells originating from the human bronchial epithelium, grown at the air-liquid interface to assess the toxicity of ordinary gasoline exhaust produced by a direct injection spark ignition engine. The transepithelial electrical resistance (TEER), production of mucin, and lactate dehydrogenase (LDH) and adenylate kinase (AK) activities were analyzed after one day and five days of exposure. The induction of double-stranded DNA breaks was measured by the detection of histone H2AX phosphorylation. Next-generation sequencing was used to analyze the modulation of expression of the relevant 370 genes. The exposure to gasoline emissions affected the integrity, as well as LDH and AK leakage in the 3D model, particularly after longer exposure periods. Mucin production was mostly decreased with the exception of longer BEAS-2B treatment, for which a significant increase was detected. DNA damage was detected after five days of exposure in the 3D model, but not in BEAS-2B cells. The expression of CYP1A1 and GSTA3 was modulated in MucilAir™ tissues after 5 days of treatment. In BEAS-2B cells, the expression of 39 mRNAs was affected after short exposure, most of them were upregulated. The five days of exposure modulated the expression of 11 genes in this cell line. In conclusion, the ordinary gasoline emissions induced a toxic response in MucilAir™. In BEAS-2B cells, the biological response was less pronounced, mostly limited to gene expression changes. 相似文献
